چکیده

From experiments with purified proteins, it has been concluded that factor Xla (FXla) is inhibited in plasma mainly by a,-antitrypsin (alAT), followed by antithrombin 111 (ATIll), Cl-inhibitor (Cllnh), and a2-antiplasmin (a2AP). However, the validity of this concept has never been studied n plasma. We established the relative contribution of different inhibitors to the inactivation of FXla in human plasma, using enzyme-linked immunosorbent assays (ELISAS) for the quantification of complexes of FXla with alAT, Cllnh, a2AP. and ATIII. We found that 47% of FXla added to plasma formed complexes with Cllnh, 24.5% with a2AP, 23.5% with alAT, and 5% with ATIII. The distribution of FXla between these

Inactivation of factor XIa in human plasma assessed by measuring factor XIa-protease inhibitor complexes: major role for C1-inhibitor.

From experiments with purified proteins, it has been concluded that factor XIa (FXIa) is inhibited in plasma mainly by alpha 1-antitrypsin (a1AT), followed by antithrombin III (ATIII), C1-inhibitor (C1Inh), and alpha 2-antiplasmin (a2AP). However, the validity of this concept has never been studied in plasma. We established the relative contribution of different inhibitors to the inactivation o...

Alpha - 1 - antitrypsin - Pittsburgh A

Alpha-l-antitrypsin-Pittsburgh is a human variant that resulted from a point mutation in the plasma protease inhibitor, a,-antitrypsin (358 Met Arg). This defect in the a,-antitrypsin molecule causes it to have greatly diminished anti-elastase activity but markedly increased antithrombin activity. In this report, we demonstrate that this variant protein also has greatly increased inhibitory act...

Regulation of Factor Xla Activity by Platelets and a 1 - Protease Inhibitor

We have studied the complex interrelationships between platelets, Factor XIa, a1-protease inhibitor and Factor IX activation. Platelets were shown to secrete an inhibitor of Factor XMa, and to protect Factor XIa from inactivation in the presence of a1-protease inhibitor and the secreted platelet inhibitor. This protection of Factor Ma did not arise from the binding of Factor XIa to platelets, t...

Characterization of a novel variant of the second domain of bikunin with increased leukocyte elastase inhibitory activity

The light chain of inter-α inhibitor, also known as bikunin or urinary trypsin inhibitor, is composed of two tandemly arranged Kunitz-type protease inhibitor domains. The second domain of bikunin has factor Xa inhibitory activity which previously was enhanced by mutating two amino acids, glutamine 19 and tyrosine 46 to lysine and aspartate, respectively. In this study, we tried to potentiate it...
